Research--Viral hepatitis and hepatocellular carcinoma; Development of urease inhibitors of gut microbiota
1. HCV Biology
The above figure shows HCV genome, infectious lipo-viral particle, and proteins involved in formation of replicase complexes (NS3, NS4A, NS4B, NS5A, NS5B). Akt1 is involved in the late stage of the HCV life cycle, that is, Akt1 maintains sufficient lipid droplet that is required for assembly and release of infectious virions. ApoE contains two Akt phosphorylation sites. We currently make an attempt to determine whether Akt regulates ApoE stability and thereby the formation of infectious lipo-viral particles.
2. Development of urease inhibitors of gut microbiota
  • Immune response in normal gut. Balanced TH17 immunity protects gut mucosa from invasion of commensal bacteria. NH3 level is low but enough to provide nitrogen to maintain homeostasis of gut microbiome.
  • Immune response in the gut with dysbiosis. Ureolytic pathogens invade the epithelial layer and stimulate DC to secrete cytokines that induce significant level of TH1/TH2 differentiation, leading to chronic inflammation. NH3 level is elevated and functions as a proinflammatory molecule that enhances TH1/TH2 activation.
